What is the MOA Cicloporin?
Binds to cyclophilin, CsA specific competitive –| of calcineurin Ca2+ & calmodulin-dependent phosphates.
What are the indications of Cliclosporin?
IBS
Immunosuppresive therapy in transplant Pt
Psoriasis
Severe atopic dermatitis
RA
What are the S/E of cicloporin?
Common:
Headache, Tremor, Hypotension, Hirsutism, renal impairment.
Other S/E:
Immunosuppresion increases risk of infections.
What are the contra-indications of cicloporin?
Abnormal baseline renal functions
Malignancy
Uncontrolled hypertension
Uncontrolled infection
What are the monitorin of ciclosporin?
Renal function
Hepatic Function
Blood pressure
Lipids
Electrolytes - Pottassium, Magnesium
Uric Acid
What interactions does ciclosporin have?
CYP450 –| : Macrolides, diltiazem, verapamil, lercidapine, fluconazole -> increase blood ciclosporin lvls.
CYP450 Inducers - Rifampicin, carbamezepine, phenobarbital, phenytoin, st jhons wort -> decrease blood cicloproin lvls.

What are the differences between IV & oral preperations of cicloporin?
Oral doses is approximatley 3x that of the IV formulation
