Drugs Used in Bone and Mineral Disorders Flashcards
Name the bisphonates and what is a common suffix for this class of drugs?
“dronate”
alendronate
pamidronate
MOA bisphosphonates.
Dir. inhibit bone resorption by inhibiting enzyme farnesyl pyrophosphate synthase (FPP)
- inhibition of FPP synthase disrupts proteins which are responsible for maintaining cytoskeletal integrity of osteoclast
*osteoclast detachment is promoted > reduced bone resorption
Indications bisphosphonates?
1st line for osteoporoses
Pagets disease
S/E of bisophosphonates.
bone mineralization defects (etidronate and pamidronate)
GI distress including esophageal ulcers (alendronate)
Consideration when taking bisphosphantes to avoid GI issues and esophageal ulcers?
Take upright and don’t take food for about 30 minutes after taking drug
MOA teriparatide?
recombinant DNA PTH analog
Indications teriparatide.
1X daily to stimulate osteoblasts and new bone formation
S/E of teriparatide use?
Risk of osteosarcoma which is increases if taken more than 2 years
MOA denosumab.
inhibits RANK ligand, a protein which acts as the primary signal for bone removal
