Dopamine Flashcards
5-HTP
Indications: Depression MoA: 5-HT precursor Adverse effects: Contraindications: Misc:
Bupirone
Indications: Anxiety, depression (off-label use)
MoA: 5-HT1A partial agonist
Adverse effects: Early increase in anxiety, drowsiness, nausea
Contraindications:
Misc:
Sumatriptan
Indications: Migraine
MoA: 5-HT1D agonist
Adverse effects: Coronary vascoconstriction
Contraindications: CAD (receptors can be found in coronary arteries)
Misc: Inhibition of inflammatory mediation release, cerebrovasoconstriction
Risperidone
Indications: Schizophrenia with psychosis
MoA: 5-HT2A/2C antagonist
Adverse effects: Weight gain/akathesia
Contraindications:
Misc: Suppresses DA release in mesolimbic (reward) pathway to decrease hallucinations and increases DA release in mesocortical pathway to stop flat affect
Odansetron
Indications: Chemotherapy-induced emesis MoA: 5-HT3 Adverse effects: Well-tolerated Contraindications: Misc:
Fluoxetine
Indications: Depression, OCD, anxiety, panic disorder, PTSD, social phobia
MoA: SSRI
Adverse effects: Sexual dysfunction, insomnia
Contraindications: MAO inhibitor (can cause serotonin syndrome)
Misc: Increase 5-HT release which initially causes self-inhibition but eventually 1a receptors desensitize and allow more serotonin release
Sertraline
Indications: Depression, OCD, anxiety, panic disorder, PTSD, social phobia
MoA: SSRI
Adverse effects: Sexual dysfunction, insomnia
Contraindications: MAO inhibitor (can cause serotonin syndrome)
Misc: Increase 5-HT release which initially causes self-inhibition but eventually 1a receptors desensitize and allow more serotonin release
L-DOPA
Indications: Parkinson's disease MoA: Dopamine precursor Adverse effects: ARRHYTHMIA, DYSKINESIA, nausea, anxiety, hallucinations Contraindications: Misc:
Bromocriptine
Indications: PARKINSON’S DISEASE
MoA: D2 agonist
Adverse effects: cardiac valvular fibrosis
Contraindications:
Misc: Post-synaptic activation in basal ganglia, neuroprotective (free radical scavenger, reduction of DA tunrover)
Carbidopa
Indications: Parkinson’s disease
MoA: Aromatic acid decarboxylase inhibitor
Adverse effects: (augment L-DOPA side effects) arrhythmia, dyskinesia
Contraindications:
Misc: Inhibits production of DA, used with L-DOPA to prevent CV effects that occurs from excessive dopamine. Helps L-DOPA to get into the brain
Selegiline
Indications: Parkinson’s disease, depression
MoA: MAO-B inhibitor
Adverse effects: HYPOTENSION (DIZZINESS), dry mouth, POTENTIAL FOR HYPERTENSIVE CRISIS/SEROTONIN SYNDROME
Contraindications: INDIRECT ACTION SYMPATHOMIMETICS or drugs that increase serotonin neurotransmission
Misc: Selective to MAOb so that MAOa can continue metabolizing tyromine and patients don’t have to worry about their dietary intake
Talcapone
Indications: Parkinson’s disease
MoA: COMT inhibitor
Adverse effects: Dyskinesia, hallucinations, nausea, hypotension
Contraindications: LIVER FAILURE
Misc: Inhibits COMT, given alongside L-DOPA
Methyphenidate
Indications: ADHD
MoA: Dopamine reuptake inhibitor
Adverse effects: Tachycardia
Contraindications: TRICYCLIC ANTIDEPRESSANT, arrhythmia, hypertension
Misc: Increases DA in the synapse which helps with focusing attention
Metaclopramide
Indications: Nausea
MoA: D2 antagonist
Adverse effects: Akathesia, focal dystonia
Contraindications: Long-term rx can cause tardive dyskinesia
Misc:
Haloperidol
Indications: Psychosis MoA: D2 antagonist Adverse effects: Extra-pyramidal motor disturbances Contraindications: Misc: Blocks DA in mesolimbic pathway
