Types of DNA libraries
A
- Whole genome (genomic library)
- Single chromosome (chromosome-specific library)
- Gene/set of genes that are expressed in a single cell type or at a certain time (cDNA library – transcriptome

To make a genomic DNA library
A
- chromosomal DNA is extracted.
- It is digested with restriction enzymes, and the resulting fragments are inserted into vectors
- Vector selection: the library ideally contains the whole genome in the smallest number of clones.
- BACs and YACs used to accommodate the large sizes of DNA necessary to span the approximately 3 billion bp of human DNA (as in the Human Genome Project).
