How does cisplatin work?

A

Cisplatin binds to DNA, and causing “kinks” in the DNA helix.

  • This stops the proteins that replicate DNA from copying properly.

And this causes tumour cells to stop reproducing.

Ligand substitution :

A

Nitrogen on one guanine base forms a dative covalent bond with cisplatin and replaces on of the chloride ion ligands.

Another nitrogen on another guanine base binds to platinum and replaces the 2nd chloride ion ligand

  • cisplatin complex bound to the DNA, strands causes the DNA strands to kink, which means that the DNA strands can’t unwind or be copied properly, so the cell cant replicate.
