Q

What are the two categorical groups of Corynebacteria?

A

Corrnebacterium Dipththeriea (casuses diphtheria)
All other Corynebacteria, called diphtheroids, which are normal inhabitants to our skin and throat.

Q

Does Diphtheria cause systemic infection?

Q

What is the source of the Diptheria toxin gene?

A

Originated in a bacteria phage. This is a case of “lysogenic conversion.” Discussed in a later class.

12
Q

Explain the endocytosis and effect of Diphtheria toxin into cells.

A

Diphtheria toxin has two domains.
Domain B binds to cell surface receptors and caused endocytosis.
Domain A is extruded from the endosome into the cell.
Domain A blocks EF2 elongation factor and hence blocks protein synthesis

13
Q

What is the initial “tentative” diagnosistic technique for Diphtheria?

A

Characteristic growth on Tellurite agar, Tinsdale’s medium, and Leoffler media.
Metachromatic granules can be visualized.

Q

Wha is the Elek test?

A

On agar, single streak of bacteria that perpendicularly bisects antitoxin paper.
A line of precipitate forms on the line Y = X
See page M-7 for details

16
Q

When shoule treatment for Diphtheria begin?

A

Begin as soon as clinical Dx is reached. Do not wait for labs to confirm.

17
Q

What is the Tx for Diphtheria?

A

Human or Horse Antitoxin
plus penicillin to kill intracellular bacteria

18
Q

What precaution should asymptomatic people take if they have had exposure to Diphtheria?

A

reimmunization as a precaution.
