2
Q

improved sensitivity

A
  • More alleles= more information
  • But improved sensitivity can lead to contamination issues
  • Pick up more background dna, can make interpretation more difficult

Q

faster results

A
  • PCR cycling time is coming down
  • 2.5 hours  50 minutes
  • Rapid DNA systems

Q

rapid DNA

A
  • IntegenX RapidHIT system
  • ~ 90 mins from sample to profile
  • Extraction, amplification, electrophoresis and analysis in one
  • Mobile system, taking analysis out of the lab
  • Profiles now being loaded to the NDNAD

Q

Higher information content

A
  • Reluctance to move away from DTRs as all current systems and databases are based on them
  • Next generation sequencing allows other markers to be analysed alongside STRs
  • mtDNA, Y-STRs, SNPs
  • STR sequence as well as length- mixture deconvolution
  • Large amount of information- bioinformatics
  • Need to work out how to incorporate and report all the new information

Q

DNA databases

A
  • By 2025 the number of reference DNA profiles in databases is predicted to ris to ~100-150 million
  • Several large countries are close to implementing nation DNA databases: Brazil, South Africa, Vietnam, Thailand, India
  • In July 2015, Kuwait announced compulsory DNA testing for all residents
  • Anyone who doesn’t comply could face a large fine and 1 year in prison

Q

cross border sharing of DNA information

A
  • Terrorism, migration, cross border crime
  • Prum convention in Europe
  • Allows searching of DNA databases between member countries
  • European standard STR loci

direct to consumer DNA testing

A
  • The rise of “recreational” DNA testing
  • Family history and ethnicity testing
  • Who owns your DNA?
  • What is it being used for?
  • Golden state killer case
