Copper Flashcards
Biochem function
component of Cu Metalloenzymes. acting as oxidases to reduce molecular oxygen
lysyl oxidase
crosslinking collagen + elastin
Cu/Zn SOD
defense against oxidative damage
Dopamine
(monooxygenase). uses Vit C, Cu, O2 to convert to norepinephrine
monoamine oxidase (MAO)
serotonin degradation, metabolism of catecholamines.
MAO inhibitors are antidepressants
Ferroxidases
oxidizes iron. Cu enzymes in plasma and function in Fe2+ oxidation needed to bind iron to transferrin
Ceruloplasmin
carries Cu in plasma
Cu deficiency
usually secondary to intestinal resection,
inc Zinc intake, or infants fed cow’s milk
Cu Absorption– transport proteins
Cu transporter 1 (CTR1)
ATPase7A
Metallothionine
(-) absorption: Zn, Vit C, fructose
Assessment
plasma Cu, serum ceruloplasmin, RBC SOD activity, platelet Cu
physiologic functions
connective tissue formation, CNS, cardiac function, cholesterol metabolism, immune competence
acrodermatitis enteropathica
autosomal recessive, low blood Zn. (MNT- Zn)
Sx: skin inflammation, diarrhea, balding, dec wound healing, immune deficiency
Menke’s disease
recessive x linked mutation in ATP7B, Cu transporting ATPase
inc Cu in liver (–> cirrhosis, liver failure), brain (–> movement disorders, seizures, personality changes, depression, psychosis, kayser fleischer rings
