What’s Hi-C? Purpose / workflow

A

Combination of 3C (chromosome conformation capture) and next-generation sequencing.

Purpose: Assay chromatin conformation. The assay detects DNA regions which physically interact with each other.

Workflow in a nutshell
1. Cross link chromatin / proteins w/ formaldehyde.
2. Chromatin is fragmented.
3. Interacting loci are ligated to a library of DNA chimeras.

What’s CLR?

A

CLR = continuous long reads.

Example : SMRT sequencing. A polymerase is attached to the bottom of a well. It reads a circular DNA and sequences it around and around.

What’s the bateson-dobzhansky-muller (BDM) model of speciation?

A
  1. Individual mutated alleles may arise in subpopulations of a species.
  2. The mutated alleles may lead to embryonic lethality or genetic diseases when combined.
  3. The subpopulation progeny will not be evolutionarily viable.
  4. The two populations stop breeding with each other.
  5. The two populations start travelling on separate evolutionary lineages.
