What’s Hi-C? Purpose / workflow
A
Combination of 3C (chromosome conformation capture) and next-generation sequencing.
Purpose: Assay chromatin conformation. The assay detects DNA regions which physically interact with each other.
Workflow in a nutshell
1. Cross link chromatin / proteins w/ formaldehyde.
2. Chromatin is fragmented.
3. Interacting loci are ligated to a library of DNA chimeras.

What’s CLR?
A
CLR = continuous long reads.
Example : SMRT sequencing. A polymerase is attached to the bottom of a well. It reads a circular DNA and sequences it around and around.

What’s the bateson-dobzhansky-muller (BDM) model of speciation?
A
- Individual mutated alleles may arise in subpopulations of a species.
- The mutated alleles may lead to embryonic lethality or genetic diseases when combined.
- The subpopulation progeny will not be evolutionarily viable.
- The two populations stop breeding with each other.
- The two populations start travelling on separate evolutionary lineages.
