CD4 and CD8 were important, needed to be a broad and high magnitude CD4 and CD8 T cell response.
Involved IFn-y production from CD8 specific T cells.
What kind of T cell response exists after HCV perstistnec and which is associated with escape?
mostly monospecific, exhuasted phenotype, but no escape from CD4 but escape from CD8.
What phenotype marker do MAIT cells in the liver have? and what TCR?
Va7.2 MAIT cells
CD161 in the liver, give very fast IL-17 responses (as well as IFN-Y).
What are MAIT cells resticted by?
Va7.2 with Ja33/12/20 and restricted to MR1.
what is presented on MR1 for MAIT cell recognition?
riboflavin, 5-OP-RU
Transcription factors of MAIT cells?
RORyt adn PLZF.
may also sense viruses.
What three viruses can activate MATI cells in vitro?
dengue, HCV and infulenza- can be independent of MR1 but rapid.
Three cytokines particuarly important in activation of MATI cells?
Il-12 Il15 and Il18.
Are MAIT cells protective in flu in mice and are they doing killing?
protective but not thorugh to be viral elimination.
Early alert system and infiltiration of T cell and macrophages is faster (overlap with ILC1 roles)
What mechanism might Ad vectors use to stimulate MAITs?
trigger pDC IFN-a. This stimulates monocyte production of IL-18 and TNF-a.
IFn-a+ TNF-a + IL-18 stimulate MAITS.
INF-a.TNF>IL-18.
