First order kinetics
Almost all drugs, clearance is constant over the concentration range, constant fraction of drug eliminated over time, elimination is not saturated, rate of elimination is proportional to the concentration
Zero order kinetics
Constant amount of the drug is eliminated over time, clearance is saturated, independent of the amount of drug present
Zero order kinetic drugs
Phenytoin, ethanol, aspirin (PEA)
Clearance (CL)
Volume of blood cleared of drug per unit time
Equation for clearance
Clearance = rate of elimination / drug concentration, clearance is in units of volume/time
Elimination
Process of drug being cleared from the system, can occur in the kidney, lungs, liver, blood, etc, clearance predicts the rate of elimination
