Chapter 2 Flashcards
How do you determine the magnification when using a microscope?
Objective x ocular
What is resolving power?
The ability to distinguish b/t two close objects
d=0.5λ/ n sin θ
Numerical aperture
What does λ stand for in the numerical aperture?
amount of light that must pass b/t two objects for them to be seen as separate
What does “n” stand for in the numerical aperture?
Light bending properties (refractile index)
What is the refractile index of air? Oil?
n Air = 1.0
n Oil = 1.5
Light microscopes
1000-2000x magnification
use immersion oil, and blue light
Different types of light microscopes?
Bright field, dark-field, phase-contrast and fluorescence
Electron microscopes
Beam of electrons in a vacuum, magnification over 100,000X, resolving power = 0.005nm
Different types of electron microscopes?
Transmission and Scanning
