Ch 5: Diagnostic Microbiology Flashcards
What does successful identification of microbe depend on?
Using aseptic technique, correctly obtaining and handling the specimen, quickly transporting the specimen to the lab
Name three methods used to identify microorganisms
Classic microbiology, molecular microbiology (genetic), immunological analysis
What are the five steps in the classic microbiological route to obtain a pure culture?
Inoculation, Incubation, Isolation, Inspection, Identification
What is Inoculation?
Introduce bacteria into a growth medium using “aseptic technique” to prevent contamination
What is Incubation?
Allow organisms to grow under the optimal conditions (temp, with or without oxygen…)
_____ is an enzyme that neutralize toxic forms of oxygen metabolites (H2O2)
Catalase
ToF: Anaerobic bacteria usually have the catalase enzyme
F
What does catalase break down?
Breakdown hydrogen peroxide (H2O2) into oxygen and water
What does the oxidase test confirm?
If the microbe is aerobic or anaerobic
How does an oxidase test work?
It identifies bacteria that produce cytochrome c oxidase, an enzyme of the bacterial electron transport chain
ToF: In an oxidase test, an anaerobic microbe will result in a colour change
F: Colour change occurs when cytochrome c oxidase present in aerobic bacteria oxidizes the agent to purple colour end product,
