CFP II Block 2 Flashcards
Basal Transcription Machinery
composed of RNA polymerase II and some general transcription factors
Cis- Regulatory Sequence
Regions of DNA that regulate the expression of genes when bound by transcription factors. Can be close to or far away, promotor, enhancer, silencer.
Co-Activator
This protein helps transcription factors to bind to the basal machinery. It is often found between activators and the basal machinery
Activator
Nuclear proteins which bind to enhancers and the basal machinery to increase transcription levels
Repressor
Nuclear proteins that bind to silencers and the basal machinery to decrease transcription levels
Promotor
This is the sequence of DNA where RNA polymerase II and general transcription factors bind to initiate transcription
Enhancer
Cis-regulatory sequence that increases transcription levels when activators bind to it
Silencer
Cis-regulatory sequence that reduces transcription levels when repressor bind to it
General Transcription Factors
Transcription factors are always found associated with RNA polymerase II when gene needs to be transcribed and is part of the basal machinery
Restriction enzymes (endonuclease)
an enzyme that cleaves (cuts) DNA at or near specific recognition nucleotide
-Type II most common
Expression Vectors
a plasmid or virus designed for protein expression in cells
ligase
joins
Terminal deoxynucleotidyl transferase
add complimentary sticky ends
