Cancer Chemotherapy Flashcards
What do alkylating groups cause
DNA cross linking and inhibit cell replication and RNA synthesis
Cyclophosphamide
Prodrug given orally or IV
“nitrogen mustard”
Lomustine
Prodrug given IV
Cisplatin
Causes renal damage and ototoxicity
Doxirubicin
Antibiotic given IV
Blocks DNA and RNA synthesis
Produces FR - heart muscle degeneration
Dantinomycin
Intercalates with DNA and interferes with mRNA synthesis
Bleomycin
Intercalates with DNA and causes its strand breakage - pulmonary fibrosis
Methotrexate
Inhibits dihydrofolate reductase - myelosuppression
5-flrorouracil
Prodrug that inhibits thymidylate synthetase - inhibits thymidine synthesis
Cytarabine
Inhibits DNA polymerase
6-mercaptopurine
Inhibits a number of enzymes needed for purine synthesis
Vincristine/ vinblastine
Given IV - binds to microtubules and blocks mitosis
Paclitaxel
Forms abnormal microtubules
Etoposide
Inhibits topoisomerase 2 - unrepairable DNA breaks
Topotecan
Inhibits topoisomerase 1
