Burkholderia (Lec4) Flashcards
Are there any similarities between structure and sequences of BPSL1594 and CNF1?
Whilst v little sequence homology, both enzymes take up a reasonably similar shape and both contain a His-Cys pair in the AS
How are CNF1 and BPSL1594 classified as toxins? ie what are their mode of actions
Draw the structure of this deamidation
Both deamidate Gln -> Glu in CNF1 this occurs in a GTPase eg Rho so that it is constitutively active in BPSL1594 occurs in an RNA helicase (eIF4A), loses its activity, translation cannot occur
What was the name of BPSL1594 changed to?
Burkholderia lethal factor 1 (BLF1)

Why does CNF1 from E. coli have these extra domains when compared to BLF1?
these additional domains allow E. coli to secrete CNF1 and for it to successfully cross the eukaryotic cell membrane to enter the cytoplasm
due to the intracelleular nature of Burkholderia it is excreted straight into the cytoplasm
