Brightfield Microscopy (Exercise 1) Flashcards
Describe the position of your hands when carrying a microscope to and from your laboratory bench.
When carrying a microscope, you must use both hands, your left hand should be under the base and your right hand holding the arm.
Why is it advisable to start first with the low-power lens when viewing a slide?
It is advisable to start with the low-power lens so you are able to find what you are looking for, once you locate what you need, center it, and you can increase magnification.
Why is it necessary to use oil in conjunction with the oil immersion lens and not with the other objectives?
The mineral oil has the same refractive index as glass, which prevents diffraction, ultimately maximizing resolution.
Oil Immersion
Objective lens provides the highest magnification, also has the shortest working distance
High-dry
This objective lens provides the second highest magnification
Low Power
This objective lens provides the lowest magnification. The coarse focus knob should be adjusted only when using this lens.
Condenser
This lens collects and focuses light from the lamp onto the specimen. A diaphragm is used to regulate light passing through this lens.
Oculars
This lens, also known as the eyepiece, often comes in pairs. Diopter adjustments can be made to this lens.
The coarse and fine focus knobs adjust the distance between:
the stage and the objective lens
A microscope that maintains focus when the objective magnification is increased is called:
Parfocal
The total magnification achieved when using a 100X oil immersion lens with 10X binocular eyepieces is:
1000X
Before the oil immersion lens is rotated into place, you should:
Center the object of interest in the preceding lens, and place a drop of oil on the slide.
