BOP2 Flashcards
What phase of trial is the BOP2 method used for
Phase II
What approach does BOP2 take
Bayesian
What does BOP2 combine
Benefits of both a Bayesian and frequentist approach
What does BOP2 stand for
Bayesian Optimal (design for) Phase 2 (clinical trials )
What does the BOP2 trial design use to make the interim decision
Probability of futility
How is the prob of futility used at the interim decisions
Compare the prob of futility to a threshold. If it exceeds the threshold, there is a higher probability that the treatment is futile than we are willing to accept.
How does the BOP2 trial design differ from other Bayesian designs
since we gain more information as the trial progresses, the first decision will be less informed than any subsequent decisions. The BOP2 method allows the threshold for probability of futility to differ between decisions, allowing thresholds to be more demanding as we gather more data. Other bayesian designs hold all decisions to the same standard.
How does the BOP2 trial design allow the threshold for different decisions to change
Define the threshold of futility as a function of the total sample size
What kind of function for the threshold for prob of futility do we use to ensure later decisions are more demanding
Monotonically decreasing
What probability of success would suggest the treatment is futile and why
0.5, suggests there is just as much chance of no response than response.
