What is hydroxyurea?
Cell cycle phase specific antineoplastic agent
Stops myeloid cell production in CML. Increases HbF in SCD, AML, and PV.
inhibits ribonucleotide reductase to stop DNA synthesis
What is imatinib/GLEEVEC?
Tyrosine kinase inhibitor that is first-line therapy for CML
What are dasatinib/nilotinib?
2nd generation tyrosine kinase inhibitors
How is CYP34A relevant to imatinib, dasatinib, and nilotinib?
CYP34A inhibitors can increase effect since all are metabolized by it.
What is ruxolitinib?
A JAK2 blocking action; Used in myelofibrosis, PV, and ET
What are sorafenib, midostaurin, and gilteritinib?
Blocks FLT3 inhibitor used for AML
What are bortezomib and carfilzomib?
Proteosome inhibitors and used in multiple myeloma and mantle cell lymphoma.
What is rituximab?
Anti-CD20 monoclonal Ab so used in large B cell CD20 positive lymphoma and NHL’s
What is alemtuzumab?
Anti-CD52 (induces lysis of both T cell and B cell populations), so is used in CLL (puts patient at increased risk for pneumo jiroveci)
What is brentuximab?
Anti-CD30 used for Hodgkins
What is epratuzumab?
Anti-CD22 used for ALL
What is daratumumab?
Anti-CD38 used for multiple myeloma
What is blinatumomab?
Anti-CD19 and anti-CD3 (bispecific Ab), being tested in ALL
What are thalidomide and lenalidonmide?
Immunomodulatory drugs that inhibit basic fibroblast growth factor and is used front line with steroids for multiple myeloma
Selectively reduces TNF-𝛂
What are pembrolizumab and nivolumab?
checkpoint inhibitors (PD-1 on T cells) with best responses in melanoma and NSCLC
