Q

What are adapter sequences?

A

contain the sequencing primer binding sites, the index sequences, and the sites that allow library fragments to attach to the flow cell lawn.

the sequencing primer anneals to the adapter, immediately upstream of the DNA insert

Q

Promoter region, transcription factors

A

Bacterial: Promoters in bacteria contain two short DNA sequences located at the -10 (10 bp 5’ or upstream) and -35 positions from the transcription start site (TSS); the Pribnow box (TATAAT) is located at the -10 position and is essential for transcription initiation (EQUIVALENT TO TATA BOX IN EUKARYOTES)
