Autonomics: Cholinomimetics Flashcards
What is the meaning behind “cholinomimetic”?
They mimic the effects of acetylcholine, the primary NT of the parasympathetic nervous system
What are the two types of acetylcholine receptors?
Muscarinic (M1, M2, M3)
Nicotinic (Smoker with a adrenal beanie)
Where are nicotinic acetylcholine receptors found?
They are found on autonomic ganglia (the transformer), on skeletal muscle motor end plates (outlet near the smoker), and in the adrenal gland (adrenal beanie on the smoker)
What is the MOA of nicotinic receptors?
Acts as ion channels that will influx positive ions to polarize a muscle
(ion channel news behind smoker)
What are the muscarinic receptors coupled to, respectively?
M1 - Gq
M2 - Gi
M3 - Gq
(QIQ store)
What is the MOA of Gq proteins?
They activate the IP3-DAG cascade leading to increased intracellular calcium
(3 dags)
What is the MOA of Gi proteins?
Decreases cAMP
packed up tent
Where are M1 receptors found?
In the nerves and CNS
the biker’s brain helmet
Where are M2 receptors found?
In the atria, the SA and AV node
the biker’s torn shirt, only top of heart is seen
Where are M3 receptors found?
On the glands
the third biker is holding a spongy gland
What is the MOA of M3 receptors?
Receptor activation leads to nitric oxide release in vascular smooth muscles, leading to increased cGMP and vasodilation
(dilated nitric oxide exhaust on the bike)
What is the meaning of the clogged exhaust pipe on the third motorcycle?
Atherosclerosis can cause vascular epithelial damage, leading to the activation of direct muscarinic receptors, causing vasoconstriction
Why do muscarinics cause a drop in blood pressure?
All muscarinics can cross to work on other M rececptors in high doses
What is Bethanechol used for?
Used to treat non-obstructive GI dysmotility
do not obstruct sign
What is the MOA of Bethanechol?
Muscarinic agonists increase secretion and motor activity of the gut
