What are the principles of autoclave operation?

A
  1. Condensation on cold surfaces creates negative pressure and draws in additional steam.
  2. Moist heat kills microorganisms by irreversible coagulation and denaturation of enzymes and structural proteins.
  3. Ideal steam for sterilization is dry saturated steam and entrained water (dryness fraction > or = 97%)
  4. Two Types:
    a. Gravity Displacement (conventional)
    b. Vacuum assisted
