It is the process of cleansing an object or substance to remove contaminants such as micro-organisms or hazardous materials.
Decontamination.
It’s primary objective is to reduce the level of microbial contamination so that infection transmission is eliminated.

It eliminates nearly all recognized pathogenic microorganisms but not necessarily all microbial forms (e.g. bacterial spores) on inanimate objects.

Factors affecting the effectiveness of a disinfection procedure:
- Nature and number of contaminating microorganisms.
- Amount of organic matter present.
- Type and condition of instruments, devices, and materials.
A process that completely eliminates all viable microorganisms, including the most resistant forms.
Sterilization
It is the most dependable system available for the decontamination of laboratory waste and the sterilization of laboratory glassware, media, and reagents.
Autoclave
It uses pressurized steam to destroy microorganisms.

What are the conditions used when sterilizing using autoclave?
Temperature: 121 C
Pressure: 15 psu
Time: Definite time based on requirement. 15 mins for equipments & 30 mins for wastes.
What are the principles of autoclave operation?
- Condensation on cold surfaces creates negative pressure and draws in additional steam.
- Moist heat kills microorganisms by irreversible coagulation and denaturation of enzymes and structural proteins.
- Ideal steam for sterilization is dry saturated steam and entrained water (dryness fraction > or = 97%)
- Two Types:
a. Gravity Displacement (conventional)
b. Vacuum assisted
What type of water should be used on autoclave?
Distilled water
Why should autoclave not be overfilled/overloaded?
There would be incomplete sterilization. Some of the materials or wastes may still be contaminated.
Why should bleach not be used when cleaning an autoclave?
Bleach (Sodium hypochlorite) is a volatile & corrosive disinfectant.
How often should a biological indicator used on autoclaves?
At least once a week
Why should you not tightly close waste bags when using autoclave?
Steam cannot penetrate the bag leaving possible contamination.
