Reversible competitive of H1
receptors
• Cross BBB and enter the CNS readily⇒ sedation
• Short duration of action (t ½ 4-6 hrs)
• E.g diphenhydramine,
chlorpheniramine and promethazine
1st generation (sedating antihistamines)

• Do not ross BBB ⇒ no sedation
• Long duration of action (t ½ 24 hrs)
• E.g Loratadine, cetirizine and
terfenadine
2nd generation (low-sedating antihistamines)
Indications:
• Allergic conjunctivitis, urticaria, angioedema.
• Adjunctive in anaphylaxis (H1antagonist, and epinephrine must also be used in anaphylaxis)
• Sedative/sleep aid.
• Antivertigo
