What is the MOA of Aspirin?
It is an irreversible inhibtor of COX and will result in no PGs being produced and no thromboxane being produced.
What are some adverse effects of aspirin?
GI effects, Pseudo-allergic reactions, renal effects (Na and H2O retention), Reyes syndrome
What is the MOA of glucocorticoids?
Glucocorticoid binds to the glucocorticoids response element and increases gene transcription of lipocortin. This will then bind to the phospholipase enzyme and prevent the production of arachidonic acid. This will reduce the production of prostaglandins.
What are some anti-inflammatory actions of CSs
Decrease the number of WBCs, increase neutrophils and inhibit inflammatory response
What is the the MOA of paracetamol?
Increased in cannabinoid receptor activation on the post synaptic membrane, causing relaxation , tranquillity and euphoria. It also inhibits PGs. And it activates descending serotoninergic pathway producing anti-nociceptic effects
What are the limits for paracetamol use?
10-15g can cause hepatotoxity
20-25g is potentially fatal. The antidote for paracetamol overdose is acetylcysteine.
