Applied Biotechnology Flashcards
CRISPR non-homologous end-joining
- once DNA is cut enzyme wants to prevent the drifting apart of the 2 DNA strands
- end binding proteins bind and crossbridge is formed
- gap repair and synthesis
CRISPR Homologous Recombination Repair
donor dna is inserted in the cut. this is now expressed
where do guide RNA and cas9 come from?
prokaryotes
Amniocentesis
take cell from the amniotic fluid
culture cells and test for disease
if there is disease hard to abort cause it is late
chorionic villus sampling
take cells from the fetus (chorionic villus)
test for disease
more invasive but faster
Testing for Tay-Sachs Disease
in vitro fert
when hexA gene is present (good) it will fluoresce
this will then be implanted into mother
Fluorescent In Situ Hybridization (FISH)
-changes in chromosome structure
- chromosome DNA denatured
- SS DNA probe with biotin is added
- fluorescent labelled avidin (binds to biotin is added)
- can now look thru microscope
allele specific oligonucleotide testing
blue/white colour change
Ashanti Desilva
- had severe combined immunodeficiency (SCID)
- non functional immune system
- WBC taken out and injected with enzyme adenosine deaminase (ADA)
- transferred back into blood stream. these replicated
