What is Amantidine?
A
Antiviral drug with anti-Parkinsonian properties

What is the MOA of Amantidine?
A
- May modulate dopaminergic function by indirectly modifying
DA synthesis, release, or uptake or on DA receptors (direct)- INHIBITS N-methyl-D-aspartate (NMDA) receptors

What are therapeutic effects of Amantidine?
A
Modest benefit for tremor, rigidity and bradykinesia (Early
onset or Mild PD Symptoms)
- Use: Reduce L-DOPA induced dyskinesias (via NMDA)

What are the adverse effects of Amantidine?
A
Primarily CNS = restlessness, depression, irritability,
insomnia, agitation, excitement, hallucinations, confusion
- Overdoses = acute toxic psychosis
- Others = headache, edema, postural hypotension, heart failure, GI disturbances.
