alpha 1 selective antagonists (-zosins… doxyzosin, tamulosin, terezosin) pharmacodynamics

A

htn- they competitively inhibit postsynaptic alpha-1 receptors, which result in vasodilation of veins and arterials, and a decrease in total peripheral resistance and blood pressure.

BPH- they competitively inhibit postsynaptic alpha-1 receptors in prostatic stromal and bladder neck tissues. This reduces the sympathetic tone-induced urethral stricture causing BPH symptoms.

A

HTN , ADHD

HTN: stimulates alpha 2 receptors in the brain stem, activating inhibitory neuron, resulting in reduced sympathetic outflow from CNS, decrease peripheral resistance, heart rate, BP

ADHD- moa unclear, regulated activity in the prefrontal cortex, responsible for emotions, attention and behaviors.

A

HTN

stimulation of the central alpha 2 receptors by methyldopa’s metabolite alpha methylnorepinephrine. produces a decrease in sympathetic outflow to the heart, kidneys and blood vessels.
