9 theme: gene expression. Biotechnology Flashcards
Why transfection is performed?
to introduce foreign genetic material into cells to manipulate or study their gene expression.
What doe we use to precipitate DNA, that it could be introduced to cell? (transfection)
we use Calcium Phosphate
What is DEAE Dextran and what is it used for?
polysaccharide, reagent used for DNA binding
What is lipofection used for?
To promote gene expression by delivering DNA into cells via lipid-mediated mechanisms.
How microinjection work?
injecting DNA with micropipette straight into nucleus or cytoplasm
What is infection?
introduction of genetic material into host cells using viruses
When Tetracycline Repressor binds to the Tet operator sites, it blocks transcription.
Is it true?
yes
During Tet-On System gene expression …
activated
During Tet-of system gene expression is…
expressed
How baculovirus work? Why its better than bacteria virus?
Baculoviruses naturally infect insect cells; The gene of interest (the gene that encodes the protein you want to express) is inserted into the baculovirus genome. high yields of recombinant protein
What are selection markers used for?
These markers enable researchers to distinguish between cells that have been successfully transformed (or transfected) and those that have not.
How does a possitive selective marker show that cell successfully incorporated the desired genetic material?
These markers typically confer resistance to a selective agent (e.g., antibiotic) or enable cells to grow under specific conditions.
