3.2 microscopes Flashcards
light microscope info
-cheap and easy to use
-light source is passed through specimen and image is reflected by glass lenses
-2D image
-resolution of 200nm
-magnifcation of x2000
transmission electron microscope info
-beam of electron passed through specimen from below, parts of specimen absorb electrons to appear dark
-flat 2D image formed
-has 0.1nm resolving power (better than SEM)
transmission electron microscope limitations
-must be a vaccuum
-specimen must be very thin
-may contain artefacts (not accurate image)
-complex staining process required
scanning electron microscope info
-beam of electron from above onto specimen
-electron are scattered and pattern allows for a 3D image to be made
-lower resolving power of 20nm
scanning electron microscope limitations
-must be a vaccuum
-may contain artefacts (not accurate image)
-complex staining process required
