11: transplantation 2 Flashcards
3 types of transplantation after 6-hydroxydopamine
nigral tissue (promotes recovery of drug induced rotational beahviour, sensorimotor deficits). medullary chromaffin cells + polymer encapsulated PC12/GDNF producing cells (only partial recovery). mouse embryonic stem cells
MPTP model: changes? what transplantations?
loss of DA cells, along with beahvioural changes (tremor, freezing, mvt difficulty). transplantation of nigral tissue, medullary chromaffin cells + polymer encapsulated PC12 cells
adrenal medullary cell transplant in PD patients: results? after you place ___, results?
first clinical trials, autologous transplantation: minimal benefits. place grafts in contact with CSF: significant improvements in motor + cogn function
adrenal medullary cell transplant in PD patients: autopsy reports? overall, worth it or not?
few or no surviving cells within the graft. high mortality + morbidity outweigh modest improvements.
transplant embryonic ventral mesencephalic cells: what open studies aka? showed what?
open label studies aka no comparison to control: grafted DA-ergic neruons can survive, mature + alleviate tremors
transplant embryonic ventral mesencephalic cells: double blind RCT showed?
improvements for younger (
transplant embryonic ventral mesencephalic cells: survival of cells? outcome?
transplanted cells survived but dystonia + dyskinesias reccured after 1st year for 15% pts. limited benefits, despite good graft survival
fluorodopa uptake in PD pts w/ or w/o transplantation: results + meaning
uptake goes down after sham surgery, but uptake higher after transplantation = the transplanted neurons are surviving
fluorodopa uptake in PD : dose dependence?
more donors per side = more uptake. still, overtime you see a decrease in uptake = still have degeneration
autopsy after transplantation: what you see?
immunological reactions : microglia. maybe this is what impairs the behavioural improvements
