Q

Describe embryonic stem cells

A
  1. pluripotency can be established
  2. good growth in culture
  3. can differentiate into different cell types with characteristics that are appropriate for that site
  4. indefinite dividing
  5. can generate a full organism

Q

where do embryonic stem cells come from

A

the blastocyst of the embryo

when inserted back in they get along well with everyone

Q

what happens if a stem cell is injected into the embryo at a later stage

A

fail to receive prior cues and form teratomas with no axis formation or segmentation

Q

Describe somatic nuclear transfer (SCeNT)

A

combines cloning methods with embryonic stem cell technology to produce custom made cells

PROCESS: nucleus is taken from the somatic cell of a patient and injected into the oocyte of a donor which replaces the nucleus (fuse)

stimulate it to divide

blastocyst is generated from hybrid oocyte and ES cells are isolated

13
Q

What are the determinants of one regulatory proteins?

A

Oct4 Sox2 Klf4 Myc

when these are inserted, they are injected into fibroblasts and form ES like cells and can differentiate

A

Type 1 diabetes

17
Q

What are the pros of umbilical cord blood?

A
  1. undifferentiated
  2. no ethical issue
  3. no genetic manipulations
18
Q

Describe the clinical applications of stem cells

A

repair of skin:

  • use cultured epidermal cells from healthy skin
  • repopulate damaged areas
  • immediate replacement of dermis from an artificial matrix
  • matrix of collagen and GAGs made into a thin membrane of silicone
  • fibroblasts and blood capillaries from the pt moves toward the artificial skin and reform the connective tissue
