Xiao Flashcards
What are the first three steps in tissue processing?
Fixation
Decalcification
Dehydration
Which part of the tissue processing is optional and what does it do to the tissue?
Decalcification
- removes calcium or lime salts from the tissues
What are the advantages and disadvantages of fresh tissue examination?
Advantages:
Protoplasmic activities such as mitosis, motion, phagocytosis, and pinocytosis can be seen.
Disadvantages:
Not permanent. Changes after death are commonly seen.
What are the signs of death during somatic death?
CNS failure
Respiratory failure
Cardiovascular failure
Fixation is also known as ______.
Preservation
What is the primary goal of fixation?
To preserve the morphologic and chemical integrity of the specimen
Give an example of an Additive solution for fixation.
Formaldehyde
Osmium tetroxide
Mercuric chloride
Give at least 5 characteristics of a good fixative
- Cheap
- Stable
- Safe to handle
- Kill the cells quickly
- Produce minimum shrinkage
- Harden the tissues properly
- Inhibits bacterial decomposition and autolysis
- Permit rapid and even tissue penetration
- The optimum osmolality for a specific tissue and fixative type
- Make cellular components insoluble
- Permit subsequent application of many staining procedures
Give at least 5 simple fixatives
- Aldehydes (Formaldehyde, Glutaraldehyde)
- Metallic Fixatives (Mercuric chloride, Chromic acid, Lead)
- Picric acid
- Acetic acid
- Acetone
- Alcohol
- Osmium tetroxide
- Heat
