Q

Is there a loci where one allele(two copies) occurs in all white flowered individuals and the other allele in all purple individuals?

A

Yes
PEAPCF1 and CD72
both are RFLPs(restroom fragment length polymorphism)

Q

Photo on pg. 14 in lecture 4. what does A in between PEAPCF1 and CD72 mean?

A

A is the flower colour locus, they do not know hat it was but knew where is was by using the Medicago plant in comparison.

Q

From th Barrel clover with bHLH-A sequence they made a ___

A

degenerate PCR primers- which is mixtures of oligonucleotides that are used in PCR to amplifie unknown DNA sequences

This techniqued worked for the study

Q

What is a BAC and how was it found in bHLH-A

A

Bacteria Artificial Chromosomes
They designed new pea-specific primers to identify the bHLH-A positivr BAC from a pea genome library

Two BACS were sequenced

Q

What was difference in the bHLH-A in white flowers?

What makes most mRNA get destroyed?

A

8 extra base pairs at an end of exon Vl
this causes a frame shift

this causes the mRNA is have a PTC(premature stop codon)

cells called nonsensemediated decay destroyes mRNA with PTC

Q

What did they do to confirm that this is the mutation that causes white flowers?

explain photo on pg 9 lecture 5

A

They added a wt version of the bHLH-A to the white flowers with a gene gun… this resulted in purple flowers

GFP is inserted to show the purple pigment
