Week 11 - Cancer Metabolism Flashcards
1
Q
How does the lactate shunt work?
A
- high energy demand
- high glycolytic flux
- NADH:NAD+ ratio increases
- lactate shunt regenerates NAD+
- allows glycolysis to continue
- inefficient but quick method to produce ATP very quickly
2
Q
What is the Warburg effect?
A
- cancer cells use the lactate shunt despite a high oxygen availability
3
Q
How is targeting the Warburg effect therapeutic?
A
- inhibiting MCT channel
- intracellular accumulation of lactic acid
- drop in pH
- cell death
- only targets cells using lactate shunt
4
Q
What is synthetic lethality?
A
- when a combination of two events results in cell death
- e.g. presence of FH and haemophilia inhibition
