Large round or Oval nucleus
Fine, homogeneous/uniform chromatin
1 to 5 nucleoli
Cytoplasm medium to dark blue with possible a few granules
Myeloblast
Slightly larger than myeloblast
Mostly same characteristics as myeloblast EXCEPT for many nonspecific/primary, Azurophilic (reddish-purple) granules
Nucleoli less prominent (1 to 3)
Cytoplasm usually blue
Promyelocyte
- Last stage capable of mitosis since it is last to have nucleoli
- Round or barely indented nucleus with one or no nucleoli
- Condensed chromatin early on, cytoplasm mostly blue, & 1 area is pinkish (“dawn of neutrophilia”)
- Later on, cytoplasm is bluish-pink, then grayish-pink still has some red-purple azurophilic granules pale pink secondary/specific granules begin, may blend in with cytoplasm
Myelocyte
- Indented, kidney-bean shaped nucleus
(Indented < 1/2 width) - Very dense chromatin
- Cytoplasm grayish-pink to pink
- Small pinkish to purple granules, may blend in with cytoplasm
Metamyelocyte
horseshoe-shaped nucleus
(Indented more than 1/2 width)
pink cytoplasm
small pinkish granules, may blend in with cytoplasm
clumpy chromatin
Band
A large cell with high N:C ratio (4:1)
Blast “myeloblast”
Prominent primary granules (azurophilic)
N:C ration of 3:1
Promyelocyte
“Dawn of neutrophilia”
- Nucleus more condensed with chromatin clumped and starting to “round up” with one flatter side
Myelocyte
Last stage capable of mitosis
Myelocyte
Kidney bean or peanut-shaped
Metamyelocyte
Large N:C ratio
Scanty cytoplasm
Cytoplasm is weakly basophilic, homogenous and clear without inclusions
Lymphocytes
Responsible in viral infections
Have abundant cytoplasm (light blue)
