What are Wake-promoting drugs (WPD)?
Narcolepsy is a rare sleep disorder (0.02- 0.06) characterised by excessive daytime sleepiness. It is possibly an autoimmune disorder caused by hypocretin deficiency
What is caffeine?
Caffeine is an adenosine receptor antagonist, blocking the A1a receptor therefore reducing the inhibitory effects of adenosine.
Dopamine synthesis
DA is synthesised from L-tyrosine in the cytoplasm of the neurons
DA is then converted into norepinephrine (NE) by DA β-decarboxylase
Amphetamines
They block the reuptake of monoamines, especially dopamine and norepinephrine transporters
Therapeutically, they are used to treat ADHD and narcolepsy
