What is a fluorescence of a molecule characterized by?
Its emission spectrum.
What makes a molecule a fluorophore?
- When a molecule efficentley absorbs a photon.
- When a molecule efficientley emitsa photon with longer wavelength.
- When a molecule the emmitted photon is within the visble light spectrum.
What type of cells are fluorscent dyes ideal for?
Fixed cells which means cells that are basically dead but their internal structures can still be used for research.
Where was the green fluorescence protein extracted from?
The Aequorea Victoria jellyfish.
What are the problems with the GFP?
It absorbed and emitted light poorly.
Why are we desperate of having that protein?
Because proteins can be genetically bond to protein of interest to tag to.
Why do cells accept this GFP?
Because its a protein, the cell can not cleave it, or reject because it not toxic.
What is emission volume called?
It is called point spread function
What does point spread function do?
It defines resolution
