UV-Vis Molecular Absorption Spectroscopy Flashcards
1
Q
What is Beer’s Law?
A
Beer’s Law relates Aborbance (and indirectly relates Transmittance) to the molar absorptivity, path length (of sample cuvette), and concentration of the analyte.
2
Q
What are the limitations of Beer’s Law?
A
- The results are only linear at low concentrations (i.e. dilute solutions).
- Molar absorptivity has a dependency on refractive index.

Q
Component considerations for UV Vis:
A
Sources: Continuous sources (Deuterium lamp for UV and Tungsten lamp for Vis)
Sample container: Quartz optic cuvette and fused silica
Detectors: Phototube, Photodiodes, CCDs.
