U1 KA 1.1 Flashcards
What is a hazard?
Something that could cause harm
What is a risk?
The likelihood that a hazard will cause harm
What is a risk assessment?
A process to determine the hazards present, their risks, and what control measures should be used
What is a linear dilution series?
When dilutions differ by an equal interval
What is a log dilution series?
When dilutions differ by a constant proportion
What does a colourimeter measure?
The absorbance of specific wavelengths of light in a coloured solution, or the turgerity of any solution
What colour should be chosen on the colourimeter?
One that the opposite to the colour of the solution, so that the light is absorbed and not reflected.
What is the sample container in a colourimeter called?
A curvette
How can the concentration or pH of an unknown solution be determined from its absorbance?
By using a standard curve
How are colourimeters calibrated?
By blanking, with a curvette filled with distilled water.
What is a centrifuge?
A device that spins samples very fast in order to separate mixtures by density
What is formed at the very bottom of a test tube that has just been centrifuged?
A pellet, composed of the densest substances in a mixture
What is the liquid at the top of a centrifuged test tube called?
The supernatent
What is gel electrophoresis?
When proteins and nucleic acids are separated by placing them in gel which then has a voltage applied to it, causing the negatively charged molecules to move toward the positive terminal
What does gel electrophoresis separate proteins and nucleic acids by?
For native: by charge, size and shape
For SDS-PAGE: just by size
