Topic 2.1.1 Cell Structure Flashcards
1
Q
What is Magnification?
A
The number of times larger an image is compared to the original object itself.
- the higher the magnification, the bigger the image is compared to the original object
2
Q
What is Resolution?
A
The ability to distinguish between objects that are very close together.
- the smaller the resolution distance (smaller distinguishable distance between two points) the higher the resolution
e.g. resolution of 0.5nm shows more detail than resolution of 200nm.
3
Q
How many CM in 1 NM?
A
1nm = 0.0000001cm (1 x 10^-7)
4
Q
What is the stain Acetic Orcein used for?
A
- Orcein is a reddish-brown dye
- orchil is a purple-blue dye.
- Orcein is used as a stain in microscopy to visualize chromosomes, elastic fibers, Hepatitis B surface antigens, and copper-associated proteins
5
Q
How many lenses does a compound light microscope have?
A
Two
6
Q
What are the two lenses that a compound light microscope has?
A
- objective lens (placed near specimen)
- eyepiece lens (specimen viewed through)
7
Q
What is the objective lens?
A
- placed near to specimen
- produces a magnified image
- objective lens configuration allows for much higher magnification and reduced chromatic aberration
8
Q
What is the eyepiece lens?
A
- specimen viewed through it
- further magnifies the already magnified image produced by the objective lens
- eyepiece lens configuration allows for much higher magnification and reduced chromatic aberration
9
Q
How are specimens illuminated in a compound light microscope?
A
- illumination usually provided by a light underneath the sample
- opaque (not transparent) specimens can be illuminated from above
10
Q
What are the four ways a sample can be prepared for examination by light microscopy?
A
- dry mount
- wet mount
- squash slides
- smear slides
11
Q
How is the dry mount technique used to prepare samples?
A
