Tissues Flashcards

1
Q

Preparation for tissue, time?

A

After death
Can not be deprived of circulation mor more than thirty minutes.
Put in plasma ass soon as the circulation is interrupted.
cut small pieces transferred to a needle to surface of a cover glass.
Large cultures, tissue cut in small pieces with sharp scissors or broader pieces with a razor.

Q

What Cristiani demonstrated about thyroid?

A

That a small pieces dies if exposed to the drying action of the air for more than ten seconds.
May me made in a drop of serum.

Q

How can be difficult to measure the area of growth or to photograph and observe the growing cells?

A

If the drop is thick the cells grows in many planes.
The cover glass is then inverted over a hollow slide os sufficient depth to prevent the drop from touching the bottom and seal with paraffin to prevent drying.

Q

Alexis Carrel and Montrose Burrows wrote about entire chicken fetus, what was about?

A

An entire chicken fetus of fifteen days, or small mammalian foetuses need to be spread in a thin layer over the surface of a large black glass plate and covered quickly with fluid of plasma.

Q

What to do as soon as the coagulation of the plasma has taken place? begin

A

The plates are are place in glass boxes with cotton sponges, soaked in water, which preserve the proper humidity. After that carefully sealed with paraffin and kept in such a position that the fluid products of the culture may drain to the bottom.

Q

During the growth, cultures can be removed from the incubator? How you can see the cultures are grown?

A

Yes, they can be removed for a few seconds…
Certain tissues as a malignant tumor or spleen that you can see without a microscope.
ON a hollow slide a new tissue of a culture appears as an opalescent area surrounding the primitive fragment - sharp edges - gray band. Whitish colour.

Q

How the fragment of the tissue appears before the growth?

A

Opaque, sharply, outline mass in the clear medium.. In the surrounding clear medium, the growing cells are easily detected. They appear form or polygonal bodies isolated or united by filaments or densely packed together.
CYTOPLASM FILLED REFRACTILE GRANULES
- NUCLEUS STANDS OUT AS A CLEAR AND HOMOGENOUS AREA
CONTAINS ONE OR MORE DARKER NUCLEOLI

Q

What is the best method of recording the morphology of the living cultures?

A

photopraph
they can change the shape
Mitotic figures have never been detected in a living culture, and they became visible only after staining the fixed specimen

Q

Cytologic study

A

Cultures fixed and stained
cover glass, culture adherent, separated from the hollow slides and immersed and corrosive sublimate, acetic acid, or formalin, or the various preparations of potassium bichromate solution

Q

What the purpose of a culture on a plate?

A

IT IS not the observation of the morphology of the cells, but the study of the dynamic changes undergone by the cells during their life outside of the organism, and the nature of their secretions.
