Thrombin-like enzymes Flashcards

1
Q

Structure of the enzyme

A
  • SVTLE ranges from 26-67 kDa
    • 232-236 amino acid residues
    • And often glycosylated
    • Typical beta fold similar to trypsin = 2 beta-barrels and each contains 6 beta strands with insertion of short beta strands and 3 alpha helices.
His57,Asp102 and Ser195 are conserved in most of the SVTLEs

Q

moa

A
  • During envenomation, the enzymes shown prominently : fibrinogenolysis, fibrinolysis, platelet aggregation, thrombosis, activation of coagulation factor etc.
    • Acts directly on fibrinogen to induce a clot by producing insoluble fibrin from fibrinogen
    • The clot is weak so it is is being removed = leading to defibrinogenated plasma
    • Does not activate factor XIII and thus no cross linking = frail clots
Remove fibrinogen in ur body = thus ,blood becomes unclottable = increase of risk of bleeding out

Q

toxin to therapeutics - thrombin like enzymes

A
  • Used as defibrinogentating agent in patients suffering from thrombosis and myopcardial infarction, peripheral vascular diseases and acute ischemia.
    • Fibrin sealant in some medical conditions
      Dissolve calcium oxide stones
