Q

Describe the flavivirus replication process

A
  • Enters via pH dependent receptor mediated endocytosis
  • +RNA genome is translated into one polyprotein which is cleaved to give viral polymerase and structural proteins
  • Virions bud into intracellular organelles and are transported to surfact via exocytic pathway

6
Q

What happens in a flavivirus infection?

A

Mosquito bites, virus initially replicates at the site.
Virus seeds itself in secondary target tissue, Macrophage, spleen, lymph….This is when you get flu-like symptoms
Secondary viremia starts, if no immune response you get yellow fever hepatitis….Dengue virus infects endothelial cells and leads to hemorrhagic feartures.

16
Q

Is there a vaccine for yelow fever

A

Yeah, live attenuated

17
Q

Hos is yellow fever spread?

A

Usually in the jungle through mosquitoes and primates. Humans become infected and take it back to urban environments where it is then spread through mosquitos.

18
Q

How does YF progress

A

Most cases are asymptomatic. For those that cause sickness, the initial infectionis flu like. Then comes a brief remission. Then comes the pain….fever, jaundice, bleeding, shock

19
Q

Treatment is YF

A

Supportive, fluids, vasoactive meds, etc…

20
Q

Bunyavirus structure

A

Enveloped, segmented, single strand, -RNA

21
Q

Three kinds of bunya

A

Rift valley fever, crimean congo,Hanta

22
Q

How is rift valley fever transmitted

A

Mosquitoes

23
Q

Crimean congo

A

ticks

24
Q

Hanta

A

rodents

25
Q

Buntavirus replication cycle

A

Endocytosed, viral polymerase carried within virion transcribes +RNA (this must be first!
This RNA is translated by host proteins

26
Q

Rift valley fever hihglights

A

causes abortion in livestock
potential bioterrorism agent
majorioty of human infection from blood or organs of dead animals

27
Q

RVFV in humans

A

Flu like at first, only progresses in 2% of pts,

28
Q

WHat organ is most affected by RVFV

A

Liver…also a nerological component like meningitis, fever, encephalitis, possible blindness

29
Q

Crimean congo highlights

A

headache, limb pain, bleeding from orifices

30
Q

Hantavirus highlights

A

Spread through rodents excretion, 1-2 weeks after exposure, fever, hemorhage, renal failure, 15% mortality rate

31
Q

Arenavirus

A

enveloped, segmented ambiscence, aerosol spread, bioterrorism

32
Q

arenavirus vaccine

A

No

33
Q

Filovirus structure

A

envelope, non seg, -RNA

34
Q

Two filoviruses?

A

Marburg and EBOLA

35
Q

Filoviruses highlights

A

Very high fatality, spread through direct contact with blood and body fluid

36
Q

Filo replication

A

binds a protein, Membrane fusion initiated by Cathepsin and low pH, brings in a viral RNA polymerase to make +RNA,

37
Q

Ebola is macrophage and monocyte trophic

A

True

38
Q

Ebola presentation

A

Bleeding from skin and orrifices, huge spleen, lesions on liver

39
Q

Treatment of Ebola

A

You’re fucked…take fluids and pray
