Discuss the operation of an autoclave
Items to be sterilized are placed in bags. An indicator is used that changes color when proper temp is reached. At the end pressure and temp will drop
List 4 basic rules for using a lab balance
Wear eye protection
Wear mask to avoid breathing in chemicals

How is distiller water made
The condensates is collected from steam created when water is boiled. This removes most of the common minerals but doesn’t remove co2 chlorine and ammonia
How is deionized water made
By passing tap water or distiller water through a resin column containing charged particles. The unwanted impurities in the water bind to these particles and are removed from the water
Explain the importance of equipment maintenance records
To monitor and record room temp and temp of water baths, fridges, and incubators
Check temp and chamber pressure record of autoclave after each use
Measure centrifuge timer and rpm every 3 months
Calibrate ph meter ain’t
Identify 5 basic containers used in the lab and explain the use of each
Bottles - reagents Test tubes Beakers - estimating volumes Graduated cylinders - for 24 hour urine Flasks - holding liquids etc

Advantages of plastic containers in the lab
Lightweight and impact and corrosion resistant unaffected by most aqueous solutions. Don’t release ions into solutions
Disadvantages of plastic containers
Some can bind and release solutes
Describe the proper care and cleaning for lab ware
Soak lab ware in disinfectant solution for 1 hour or overnight. Following decontamination they should be rinsed free of disinfectant and then washed for routine cleaning
List precautions when using lab ware
If damaged or chipped glassware do not use
appropriate PPE must be worn
Explain the use of a volumetric pipet
Used by attaching a auctioning aid or pipet filler to the top of the pipet. The liquid is suctioned into the pipet to slightly above the etched line. The liquid is carefully lowered until the meniscus is even with the line. The outside of the pipet is wiped. The pipet is held nearly vertical and tip is placed against the inner wall of container. Suction is released and the liquid is allowed to flow. A small drop remains in the pipet tip
Explain the use of a graduated pipet
A auctioning decide is attached. The liquid is suctioned up to just above the desired mark and then the level is lower to the desire volume. Pipet tip is wiped dry. The liquid is held almost vertically m. The last drops are forced out using the pipetting aid

Explain the use of the coarse and fine adjustments
The coarse adjustment is used to focus the lower power objective only. The fine adjustment is used to give a sharper image after the object is brought into view with the coarse adjustment
