Masson Trichrome
blue - collagen stains blue or green, cytoplasm and muscle red, nuclei are dark blue
To distinguish cells from conjunctive tissue
Will color fibrous/connective tissue in blue
Giemsa
black
Elastic van gieson
Purple/red
Elastic red picro
collagen and elastic fibers stain red and black
H&E
collagen may be unstained. Lipid won’t take up stain
* Do not color in blue Myocardial cells are pink
Periodic acid schiff (PAS)
schiff reagent targets aldehyde groupes, resulting in staining of carbohydrates. Magenta satin taken up by glycogen, mucous.
Alcian blue
neutral and acid mucopolysaccharide stain purple and blue. stains proteoglycans.
Oil red O
stains for lipid.
Lugol’s
conduction system b/c high glycogen content
Elastic van Gieson (aka Verhoeff-van Gieson)
myocytes are yellow, fibrous tissue is purple, fat is white. Elastic fibers black
Elastic red picro stain
collagen red, elastic fibers black
Stroma
= connective tissue
Is collagen, fibroblasts, macrophages, etc.
Abnormal fibrosis can be present endocardially, interstitially, or perivascularly.
Type I collagen is the major fibrillar collagen infibrous tissue.
Parenchyma
= cardiomyocytes
Heart failure cells
= macrophages laden with hemosiderin or RBCs.
Chromogranin and synaptopysin staining
= neuroendocrine tumors.
