Staining Techniques Flashcards
Most common staining method.
H&E
commonly used to stain elastic fibers dark blue to black
Weigert method and Van Gieson Stain
Stains glycogen, glycoproteins, mucins, and glycolipids
Periodic Acid-Schiff Reaction (PAS)
uses silver salts in solution that precipitate out as a silver metal on surfaces of type III collagen fibers (reticular fibers), staining them black
Silver stain
specific for staining mucins, glycoproteins and the matrix of cartilage blue in color whereas the cytoplasm stains a light pink and nuclei stain red
Alcian Blue
uses silver stains that become reduced to demonstrate calcification and calcified tissues that stain black
von Kossa stain
used to stain lipids, phospholipids, lipoproteins and triglycerides, all of which stain an intense red
Sudan Red
used to localize mucin w/c stains a deep red color
Mucicarmine stain
stains collagen blue in tissue section
methyl blue/cotton blue
used to localize granules of mast cells, cartilage matrix, and mucin of goblet cells, all of which stain orange to red
Safranin-O
metachromatic stain in that it changes color w/ specific substances such as the granules of mast cells and cartilage matrix, both of which stain a reddish-purple color.
Toluidine blue
used to detect the presence of iron; stains blue or purple
prussian blue
