Signalling through proteolysis (4) Flashcards
what is Wnt signalling ?
signalling through a ligand called Wnt (secreted protein)
what does Wnt signal through ?
canonical pathway -
planar cell polarity (PCP) pathway
Ca2+ pathway
Canonical Wnt signalling.. what is Wnt
stimulus
Frizzled and Lrp5/6
receptor
Dishevelled
Effector
B-catenin
Transcription factor
What happens when there’s no Wnt ?
B-catenin undergoes proteolysis
B-catenin forms a complex with 1.
1.Axin
B-catenin forms a complex with 2.
- Adenomatous Polyposis Coli
B-catenin forms a complex with 3.
- Glycogen synthase kinase 3
B-catenin forms a complex with 4.
Casein Kinase 1alpha
B-catenin forms this complex which undergoes ?
phosphorylation and proteolysis
At rest what is concentration B-catenin ?
very low
At rest, what stops the transcription of the genes that Wnt will eventually cause to to be expressed ?
Groucho
What prevents B-catenin proteolysis ?
Wnt
How does Wnt prevent proteolysis ?
by preventing B-catenin proteolysis - when Wnt binds to frizzled dishevelled is recruited which steals Axin therefore B-catenin proteolysis doesn’t occur.
What ultimately occurs when Wnt binds to frizzled ?
B-catenin levels increase which is a transcription factor
Why is Wnt signalling important ?
Crucial for embryonic development and nerve circuits
Hedgehog Signalling.. what are they and how many types are there ?
Secreted proteins
Three types: sonic, indian, desert.
What is Hedgehog ?
Stimulus
Patched ?
Receptor
Smoothened
Effector
Cubitus interruptus/Gli (mammalian)
transcription factor
what does cubits interrupts undergo at rest ?
proteolysis
