sem B - drug treatment of depression Flashcards
what was the 1st antidepressant drug
iproniazid
how was the first antidepressant drug discovered
new treatment for TB but cured depression - by blocking enzyme monoamine oxidase so monamines are not broken down
what is the monoamine hypothesis of depression
reduced levels of monoamine in brain causes depression (reduced noradrenaline, dopamine, serotonin)
MAO neurotransmission
-MAO neurotransmitter is packaged into vesicles and released into synaptic cleft by exocytosis
-NT activates post-synaptic MAO receptors
-MAO transporter reuptakes NT into presynaptic NT
-NT is either repackaged into vesicles or broken down by MAO into inactive metabolites
MAO inhibitors - blocks breakdown of
monoamines
tricyclic antidepressants block …… preventing reuptake into presynaptic neurone, so it builds up in synaptic cleft. are selective/non-selective
MA reuptake transporter
-selective
SNRIs - selective to …. and …. blocking their reuptake
noradrenaline and serotonin
SSRIs - selective to ….. blocking their reuptake
serotonin
NRIs - selective to …. blocking their reuptake
noradrenaline
