RP11:test diabets/glucose In Urine.🥼 Flashcards
Give me the test for diabetes with urine samples given to you.
1-Take a pair of goggles and a test tube rack with 4-6 test tubes
2-place test tube in the racks and name them accordingly A,B,C,D,E,F
And write initials
3-use different pipette to add 2cm cubed to each corresponding test tube
4-use different puppets to add 10 drops of HCI to earth test tube
5-use test tube holder to place them into water water bath of 90 degrees for 4-5 minutes.
6-after cool down test tube with cold tap water leave to cool
7-sodium hydrogenate carbonate (NaHCO3) powder until it stops fizzing
Can confirm the solution is alkaline by placing universal indicator in swells in Pasteur pipette
8-add 0.5cm3 of Benedict solution with pipette to test tube In each solution
9-heat the test solution to boiling point in water baths and heat for 2 minutes
10-observe the colour change over the 2 minutes
Why do we add HCI?
To break glycosidic bond
To release galactose and glucose in sucrose and all monosaccharides which Benedict can react with
Why do we use NaOH3 in solution?
So solution is alkaline because acids and acids don’t react but just get together ,it is needed so that Benedict reagent to work
Give me the Hazard assessment.
(Urine)
Benedict reagent:irritate skin and eyes so we must wear goggles
-And if you got any on you wash eye /skin immediately with cold water
-Low risk
-hot liquid:water bath, can cause scalding so use tongs to remove the test tube rack
-And if any hot liquid got on you just rinse affected area on cold tap water.
How to deter by colour the quantity of glucose in urine
Green:transable
Yellow:low
Orange:moderate
Brick red:high
