What is a revertant mutant?

A

A mutant with two mutations such that one mutation reverses the effect of the other, restoring the phenotype to pseudo-wildtype.

What is a suppressor mutant?

A

A mutant with two mutations that counteracts the first mutation by protein interaction.

A

Determining chemical compound mutagenicity.

How does it work?

A

Two cell plates with histidine-requiring Salmonella, on minimal media.

On one plate, a filter disk with buffer is applied. On the other, a filter disk with the testing chemical.

Random revertants will be visible on the ‘buffer plate’.

The number of revertants on the ‘chemical plate’ gives an indication of it’s mutagenicity.
